I want to change single notes whilst the sequencer is playing my sequence to try different variations on melody

can somebody point me in the right direction

Put the sequencer in step edit mode, hold the BANK button, and press a step button. Now any key you hit on the keyboard will change the note for that step the next time it plays. See page 18 (STEP EDIT MODE -> EDITING A STEP) in the manual for complete details.
