Put the sequencer in step edit mode, hold the BANK button, and press a step button. Now any key you hit on the keyboard will change the note for that step the next time it plays. See page 18 (STEP EDIT MODE -> EDITING A STEP) in the manual for complete details.

Assuming the Subsequent37 firmware is similar to that in the Sub37, I'd recommend checking the PRESET MWHL setting (item 5 on page 2 of the GLOBAL menu.) Can't help with your sync problem (Issue 1). Your Issue 2 sounds like the POLY NUMBER setting has been set to something other than OFF. One other thing you could try is to set the pattern to 'Rec' to start recording a sequence, and then pressing a key to clear the sequence. Yeah, mine does it, too, and I LOVE it! I get some great effects by setting the feedback control just slightly before the wildness starts and then modulating the wave shape with an lfo, velocity, or pressure.
I definitely see it as a feature, not a defect.

KB PRIORITY determines how the oscillators react to key presses. You're not hearing it with that patch because that patch does not retrigger the envelopes when a new key is pressed while another key is held, and the amp envelope has a quick decay and no sustain.
